If I set up multiple activities within one event that each require registration, are classmates able to return and add or delete a registration of an activity that was not selected initially?

They can add new registrations by registering another time (for a guest I presume or in the case where they are paying for another classmate after they have already registered for themselves), however, they would need to contact you to delete a registration. They can edit their answers to custom questions, however, it is not common to allow members to delete a registration without contacting you as admins are often having to submit numbers to vendors etc. and they need to be involved in the process of any deletions.

I imagine that many will register for the main reunion but may not know if they will be attending the other activities until later. I want to encourage early registration for the main event which is the only one we need to collect money. Registrations for the other activities mainly is to pre-print nametags.
So, when you say they can "register another time," they don't re-register for the main event, do they?

Yes they can only choose only one or more of the activities and register just for those. They will see all activities and they can choose a quantity for each each time they register which means they could register for 1 or more on an initial registration and 1 or more of the others (or a new one of the same one if they need to add a guest after the fact).

Once they register the top of the registration page will show all of their prior registrations at the top of the page - they can view what they ordered in case they want to print it and they can edit custom questions if there were any. As I mentioned above they can not edit quantity there. They would need to register new ones for guests or contact you if they need to delete.
